How It Works:
Vonoprazan is a potassium-competitive acid blocker (P-CAB) that inhibits H+, K+-ATPase in a reversible and potassium-competitive manner. Unlike traditional proton pump inhibitors, Vonoprazan does not need to be activated by acid. Its strong basicity allows it to remain bound to the acid production sites on gastric parietal cells for an extended period, effectively inhibiting gastric acid production. This results in significant protection against mucosal damage in the upper gastrointestinal tract.

Common: Diarrhea, constipation, nausea, abdominal distension
Less Common: Drug hypersensitivity (including anaphylactic shock), drug eruption, urticaria, hepatotoxicity, jaundice, rash
Laboratory Abnormalities: Increased gamma-glutamyl transferase, AST, ALT, ALP, LDH, and eosinophilia
Other: Edema

Indication:
Vonoprazan is indicated for:
Gastric ulcer
Duodenal ulcer
Reflux esophagitis
Prevention of recurrence of gastric or duodenal ulcers during low-dose aspirin administration
Prevention of recurrence of gastric or duodenal ulcers during non-steroidal anti-inflammatory drug (NSAID) administration
When Not to Use:
Vonoprazan is contraindicated in:
Patients with hypersensitivity to Vonoprazan or any excipient in the product
Patients receiving atazanavir sulfate, nelfinavir, or rilpivirine hydrochloride

Precaution:
Monitor the course of the disease closely during treatment. Use the minimum effective dose based on the condition of the disease.
Scivono Tablets Warnings:
Warning 1:
For maintaining healing of reflux esophagitis, Vonoprazan should be used only in patients with recurrent or persistent symptoms. Avoid use in patients who do not require ongoing maintenance therapy.
Warning 2:
If healing is maintained over a long period and there is no risk of recurrence, consider reducing the dose from 20 mg to 10 mg or discontinuing the medication.
Warning 3:
Administer Vonoprazan with caution in patients with renal disorders, as delayed excretion may increase the drug’s concentration in the blood.
