How It Works:
Ondansetron is a selective antagonist of the serotonin 5-HT3 receptor. It is believed to mitigate chemotherapy-induced nausea and vomiting by blocking the initiation of the vomiting reflex that is triggered by serotonin release from enterochromaffin cells in the small intestine. By antagonizing the 5-HT3 receptors on both peripheral and central nervous system neurons, Ondansetron helps to prevent the central and peripheral pathways involved in nausea and vomiting. The exact mechanisms of its antiemetic effect in postoperative nausea and vomiting remain unclear.

Side Effects:
Common: Diarrhea, headache, fever, constipation, rash
Cardiovascular: Rare cases of angina, ECG changes, hypotension, tachycardia, QT prolongation, ST segment depression, bradycardia
Hepatic: Transient elevations in AST and ALT, typically exceeding twice the upper limit of normal in about 5% of patients, not related to dose or duration of therapy
Neurological: Rare cases of extrapyramidal reactions, grand mal seizures
Other: Hypokalemia, headache, drowsiness/sedation, injection site reactions, fever, cold sensation, pruritus, paresthesia
Local Reactions: Pain, redness, burning at the injection site
Lower Respiratory: Hiccups
Skin: Urticaria, Stevens-Johnson syndrome, toxic epidermal necrolysis
Eye Disorders: Transient blindness
Neurological: Oculogyric crisis and other dystonic reactions, transient dizziness

Indication:
Prevention of nausea and vomiting associated with initial and repeat courses of emetogenic cancer chemotherapy
When Not to Use:
Contraindicated in patients with known hypersensitivity to Ondansetron or any of its components
Contraindicated with apomorphine due to reports of profound hypotension and loss of consciousness
Seroblock Injection Precautions:
Precaution:
Hypersensitivity reactions, including anaphylaxis and bronchospasm, have been reported in patients with previous hypersensitivity to other selective 5-HT3 receptor antagonists.
Seroblock Injection Warnings:
Warning 1:
Ondansetron can prolong the QT interval in a dose-dependent manner. Avoid use in patients with congenital long QT syndrome. ECG monitoring is recommended for patients with electrolyte abnormalities (e.g., hypokalemia or hypomagnesemia), congestive heart failure, bradyarrhythmias, or those taking other QT-prolonging medications.
Warning 2:
Ondansetron may mask progressive ileus and gastric distention following abdominal surgery or chemotherapy-induced nausea and vomiting.
Warning 3:
Ondansetron does not stimulate gastric or intestinal peristalsis and should not be used as a substitute for nasogastric suction.
