How It Works:
Peridone’s antiemetic effect is attributed to its peripheral (gastrokinetic) actions and its antagonism of dopamine receptors in the chemoreceptor trigger zone, located outside the blood-brain barrier in the area postrema.

Common: Headache (5.6%), Diarrhea (5.2%), Breast Enlargement/Gynaecomastia (5.3%), Breast Tenderness (4.4%), Galactorrhea (3.3%), Amenorrhea (2.9%)
Less Common: Depression (2.5%), Somnolence (2.5%), Rash (2.8%), Pruritus (1.7%), Akathisia (1%), Libido Decreased/Loss of Libido, Menstruation Irregular (2.0%), Lactation Disorder (1.6%)
Drug Interactions:

When Not to Use:
Contraindicated in patients with moderate to severe hepatic impairment, existing cardiac conduction interval prolongation (particularly QTc), significant electrolyte disturbances, or underlying cardiac diseases (e.g., congestive heart failure).
Peridone Tablets Precautions:
Precaution: Epidemiological studies suggest an increased risk of serious ventricular arrhythmias or sudden cardiac death associated with domeperidone.
Peridone Tablets Warnings:
Warning 1: Patients over 60 years old should consult a physician before using domeperidone.
Warning 2: Not recommended for patients with known QTc interval prolongation or significant electrolyte imbalances due to an increased risk of ventricular arrhythmia.
Warning 3: Discontinue use if signs or symptoms of cardiac arrhythmia occur and consult a physician immediately.
