How it Works:
Mesalazine exerts its anti-inflammatory effects by inhibiting the migration of polymorphonuclear leukocytes and the lipoxygenase enzyme in the large intestine. It decreases the production of pro-inflammatory leukotrienes, activates PPAR-Y receptors, and mitigates intestinal inflammatory responses.
Pentasa Tablets Usage and Safety:
Dosage: Mesalazine
Side Effects: Common: Headache, diarrhea, abdominal pain, nausea, vomiting, rash, drug fever, dizziness, arthralgia, and dyspepsia. Rare: Myocarditis, pericarditis, anemia, tinnitus, paresthesia, flatulence, pruritus, urticaria, myalgia, interstitial nephritis, pancreatitis, abnormal renal function, leucopenia, thrombocytopenia, agranulocytosis, and neutropenia.
Drug Interactions: Digoxin, Azathioprine, 6-mercaptopurine, furosemide, spironolactone, warfarin, glucocorticoids, NSAIDs, methotrexate.
Indication: Treatment of mild to moderate acute exacerbations of ulcerative colitis, maintenance of remission of ulcerative colitis, and Crohn’s ileocolitis.
When Not to Use: Contraindicated in patients with a history of sensitivity to salicylates, severe renal or hepatic insufficiency, children under 2 years of age, and with concomitant use of lactulose or similar preparations.
Pentasa Tablets Precautions:
Precaution: Renal function should be monitored with serum creatinine levels before starting treatment, every three months for the first year, then every six months for the next four years, and annually thereafter. Discontinue if renal function deteriorates.
Pentasa Tablets Warnings:
Warning 1: Serious blood dyscrasias have been reported rarely; discontinue treatment if unexplained bleeding, bruising, purpura, anemia, persistent fever, or sore throat occur and conduct hematological investigations.
Warning 2: Begin treatment with caution in patients with existing gastric or duodenal ulcers.
Warning 3: Use with caution in patients with a history of myopericarditis of allergic origin.
