How It Works: Mesalazine exerts its anti-inflammatory effects through mechanisms not fully understood. It inhibits the migration of polymorphonuclear leukocytes and the activity of lipoxygenase in the large intestine. This reduces the production of pro-inflammatory leukotrienes (LTB4 and 5-HETE) and activates PPAR-γ receptors, which help counteract inflammatory responses in the intestine.

Dosage: Mesalazine
Side Effects:
Common: Headache, diarrhea, abdominal pain, nausea, vomiting, rash, drug fever, dizziness, arthralgia, dyspepsia.
Rare: Myocarditis, pericarditis, anemia, tinnitus, paresthesia, flatulence, pruritus, urticaria, myalgia, interstitial nephritis, pancreatitis, abnormal renal function, leucopenia, thrombocytopenia, agranulocytosis, neutropenia.
Drug Interactions: May interact with digoxin, azathioprine, 6-mercaptopurine, furosemide, spironolactone, warfarin, glucocorticoids, NSAIDs, and methotrexate.
Indication: Used for treating mild to moderate acute exacerbations of ulcerative colitis, maintaining remission of ulcerative colitis, and maintaining remission of Crohn’s ileo-colitis.
When Not to Use: Contraindicated in patients with a history of sensitivity to salicylates, severe renal or hepatic insufficiency, children under 2 years of age, and those using lactulose or similar preparations.
Masacol Tablets Precautions:
Precaution: Renal function should be monitored regularly. Serum creatinine levels should be measured before starting treatment, every three months for the first year, then every six months for the next four years, and annually thereafter. Discontinue use if renal function deteriorates.
Masacol Tablets Warnings:
Warning 1: Serious blood dyscrasias, such as unexplained bleeding, bruising, purpura, anemia, persistent fever, or sore throat, have been reported rarely. Perform hematological investigations if these symptoms occur and discontinue treatment if blood dyscrasia is suspected.
Warning 2: Use with caution in patients with existing gastric or duodenal ulcers.
Warning 3: Exercise caution in patients with a history of myopericarditis of an allergic nature, regardless of its origin.
